You can take a cannon character and alter them in any way you wish. For example, Spiderman didn't get bit by a radioactive spider but rather he was given the Venom symbiote by Madame Webb that grants him powers in this game. The only characters off limits are the core Avengers so there's no limit to who you could use and/or alter.
