Ok, what about Killer Kroc starting out as a heroic cop who finds out he has a terminal illness, similar (spoiler of ASM2) to Harry Osborn? As it slowly takes over his body and mind, he starts falling into a villianous role. So a Walking The Line type character.

Or if you need an outright hero, I could try for Aquaman :)
