You mean 2 questions. :P 

The characters wont notice the changes right away thus the five day time skip then the characters will start to notice the changes, higher strength, better eyesight, hearing, scent, and healing, ect.

And the second question, yes you will be tougher to kill without silver or wolfsbane, but you can still die if enough damage is taken but wolfsbane will weaken the werewolf if it gets into their bodies somehow, and silver will kill a wolf if the wound is deep enough.
