Okay two things

1: Isa was in fact right next to Vlad.

2: Tranquilizer isn't magic, it takes several MINUTES to knock a creature out, it isn't like the movies where it knocks them out instantly. Even IF Isa is immediately shot with tranquilizer darts as soon as he starts shifting, He'd still have time to spin away from Lucifer and jump on a prone Vlad, though he would be well on his way to taking a nap once there.
