I'm honestly not even sure what "call[ing] upon the deeper nature of your powers in your sanctuary" even means. ^//^ But no; it specifically states "Once you have taken all five doomsigns above the line, you must take “Your doom arrives” the next time your doom track fills", implying you have to take all five. Dunno why you'd want to do that, though; all of them can be easily explained with your particular doom.
