So, in some respects, the plot (insofar as you will drive it) will be shaped around the player characters? Would it then be worth players, instead of submitting full sheets now, submitting a more general description of the kind of character they'd like to play? I have a medical character in mind, but I'm finding it quite tricky to put more precise details to him beyond vague background and general demeanour without knowing quite what plots he has to look forward to :P
