Well, the Raimi trilogy gave us not just the definitive J. Jonah Jameson, as stated above, but also the definitive Aunt May in the form of Rosemary Harris. (And Cliff Robertson's Uncle Ben, for that matter.) Josh Keaton's voice just is Spider-Man to me now.
I agree with both Triple J and Aunt May but I think I enjoyed Martin Sheen as Uncle Ben more. Likewise, I enjoy Michael Clarke Duncan's Kingpin more than I do Vincent D'Onofrio. Batman for me, I guess it's got to be Christian Bale (I am not including voice performances), but I do like the way Affleck's fights more so than any of his predecessors. Alfred is also a tough choice because I can't really think of a bad live-action version, Gough, Caine, Pertwee and Irons all have their merits. Though for all the things I could say poor of Gotham, I really do like Sean Pertwee's Alfred. Superman is tough because I really want to give it to Welling but I think I'm going to give it to Reeves if only because by Welling's own admission, he never played Superman, he played Clark Kent for ten years. Lois though is fully going to Erica Durance which I realize might be a less popular choice in this crowd.
