No, the main idea is that James used this capsule for another reason. It has a timer. He knows exactly when it goes off. If necessary, he can apply the solution, because he is aware of when it happens.
Mostly, I'm using it as a mechanic to both show off how ruthless James is, and to force Hotarubi into the lesson, as James would. However, would the worst come to the worst, James would be forced to save Hotarubi due to the restrictions placed upon him by Third.
Sorry, I just realized you were referring to mithridatism, and not a misspelling of metabolism.

Mithridatism is the gradual increase of a resistance against a poison by taking gradually increasing doses of that poison. Unfortunately, this would not be effective against Halon, partially because you cannot build up an immunity against the lack of oxygen. Furthermore, to say you cannot be poisoned because of mithridatism would mean that you have applied a technique that can take many years to every possible poison, which is impossible, seeing as how there are thousands upon thousands of poisons in the world, and this technique cannot be used to combat some of them.

What? James knows his poisons. He wouldn't use anything that wasn't completely and utterly lethal. Furthermore, being a small capsule, it is untraceable, has no taste, cannot be removed without extensive surgery, and is thus incredibly hard to counter.
I'm going to assume you meant to say "Due to my metabolism".
Now, the poison used by James is a substance known as Halon, in a capsule, which is ingested orally. Halon is a gas which bonds with oxygen atoms, and pulls them into itself incredibly well. Once the capsule opens, it will effectively suck all oxygen out of your bloodstream, removing your ability to absorb oxygen. It is rather like carbon-monoxide poisoning, except far more effective. Unless your metabolism allows you to survive without the need for oxygen, Halon will most definitely kill you.
At first, you will feel drowsy. It will not feel like suffocation, because you are still breathing in air. However, after around 3 minutes the brain will shut down, knocking you out. After this has occurred, it will be about 2-3 more minutes before complete brain death. However, 1 minute will be sufficient time for serious and permanent brain damage to occur, which can result in personality change, memory loss, blindness, deafness, loss of speech, loss of bowel control, paralyss in it's many forms, the loss of the ability to make new memories, dementia, and going into a coma.
All in all, it is a relatively slow-working orally ingested poison, which kills through removal of oxygen from the bloodstream.

Sighing, Bob starts turning the toilet into a hangar, before he remembers that he can't replicate the ship. Cursing his horrible sense of direction, Bob floats off to a meeting room, where, with much extravaganza, he starts his quest to find the garage. And possibly get sidetracked. That woman DOES look unhappy, thinks Bob, as he decides to use the catnip trick agan, to a far lesser effect this time.
