I haven't seen anyone accidentally mistake 'your' and 'you're' in a long time. I should change my signature to a different, more common mistake.

'Know' was probably just a lapse of attention (I'd hope), but 'affect' was more likely a genuine grammatical error. The word 'affect' is a verb. You meant to use the noun form, 'effect'.
